ASC30 is an investigational once-daily tablet from Ascletis Pharma that activates the GLP-1 receptor, the target of semaglutide. In a 13-week US phase 2 study of 125 adults with obesity or overweight and no diabetes, average weight loss beyond placebo was 5.4% on 20 mg, 7.0% on 40 mg and 7.7% on 60 mg, and 4.8% of people stopped because of side effects, all of them stomach related. In a 13-week phase 2 study of 100 adults with type 2 diabetes, HbA1c fell by 1.7%, 1.6% and 1.7% beyond placebo on 40 mg, 60 mg and 80 mg at week 12, and weight fell by 4.4%, 3.8% and 4.0% beyond placebo at week 13.
Phase 3 began dosing on August 30, 2026. The two trials, AURORA-1 (adults with obesity or overweight and no diabetes) and AURORA-2 (adults with type 2 diabetes), plan about 4,600 people in total, assigned to 20 mg, 40 mg or 60 mg or placebo for 72 weeks. Ascletis expects topline data in the third quarter of 2028 and plans a US filing by the end of 2028.
Worth knowing: These are company-reported topline results, not peer reviewed. Phase 2 stepped the dose up weekly from 1 mg, and the company plans four-weekly steps in phase 3. Its claim of less vomiting than orforglipron compares separate studies and is not a head-to-head trial. ASC30 is not approved.
